=== Phenol–chloroform extraction === The basic principle of the phenol-chloroform extraction is that DNA and RNA are relatively insoluble in phenol and chloroform, while other cellular components are relatively soluble in these solvents. The addition of a phenol/chloroform mixture will dissolve protein and lipid contaminants, leaving the nucleic acids in the aqueous phase. It also denatures proteins, like DNase, which is especially important if the plasmids are to be used for enzyme digestion. Otherwise, smearing may occur in enzyme restricted form of plasmid DNA.
A glucoside is a glycoside that is chemically derived from glucose. Glucosides are common in plants, but rare in animals. Glucose is produced when a glucoside is hydrolysed by purely chemical means, or decomposed by fermentation or enzymes. The name was originally given to plant products of this nature, in which the other part of the molecule was, in the greater number of cases, an aromatic aldehydic or phenolic compound (exceptions are Jinigrin and Jalapin or Scammonin). The simplest glucosides are the alkyl ethers which have been obtained by reacting hydrochloric acid on alcoholic glucose solutions. A better method of preparation is to dissolve solid anhydrous glucose in methanol containing hydrochloric acid. A mixture of alpha- and beta-methylglucoside results. == Structure == Cyclotides have a well-defined three-dimensional structure due to their interlocking disulfide bonds and cyclic peptide backbone. Backbone loops and selected residues are labeled on the structure to help orientation. The amino acid sequence (single-letter amino acid representation) for this peptide is indicated on the sequence diagram to the right. One of the interesting features of cyclic peptides is that knowledge of the peptide sequence does not reveal the ancestral head and tail; knowledge of the gene sequence is required for this. In the case of kalata B1 the indicated glycine (G) and asparagine (N) amino acids are the terminal residues that are linked in a peptide bond to cyclize the peptide.

== Risks == Administration of ACAM2000 poses risks and may cause side effects. Most people who have taken the vaccine only report mild reactions. Reactions may include a sore arm, fever, and body aches. Some people may have more serious side effects, including effects that may be life-threatening. According to the FDA-approved prescribing information leaflet, "Common adverse events include inoculation site signs and symptoms, lymphadenitis, and constitutional symptoms, such as malaise, fatigue, fever, myalgia, and headache." These reactions are less frequent in people being revaccinated than those receiving the vaccine for the first time. No known contraindications exist to receiving the vaccine in case of an outbreak emergency. Furthermore, it is recommended that the vaccine should be given to pregnant women who have been exposed to smallpox. "Because the risk of maternal serious illness or death, prematurity, miscarriage, or stillbirth from a smallpox infection are greater than the risk of the vaccination, smallpox vaccine is recommended and should be offered to pregnant women in case of an outbreak emergency."

Frederick, the designer of Army Special Unit knife V-42 stiletto which was based on Fairbairn–Sykes commando knife. Fairbairn also a friend of Captain Dermot Michael "Pat" O'Neill, the First Special Service Force's close-combat instructor. In 1945, Colonel Movlid Visaitov, commander of the 255th Separate Chechen-Ingush Cavalry Regiment and the 28th Guards Regiment. Visaitov was the first Soviet officer to shake hands with General Bolling at the Elbe River. On 27 September 1945, General Alois Liška of the Czechoslovak Army under former U.S. command, was awarded by General Ernest N. Harmon in Prague Group Captain Harry Day, Senior Officer at numerous POW camps during World War II, and significantly helped American POWs endure the captivity, as well as organizing escape operations. Much of the universe's water is produced as a byproduct of star formation. The formation of stars is accompanied by a strong outward wind of gas and dust. When this outflow of material eventually impacts the surrounding gas, the shock waves that are created compress and heat the gas. The water observed is quickly produced in this warm dense gas. On 22 July 2011, a report described the discovery of a gigantic cloud of water vapor containing "140 trillion times more water than all of Earth's oceans combined" around a quasar located 12 billion light years from Earth. According to the researchers, the "discovery shows that water has been prevalent in the universe for nearly its entire existence". Water has been detected in interstellar clouds within the Milky Way. Water probably exists in abundance in other galaxies, too, because its components, hydrogen, and oxygen, are among the most abundant elements in the universe. Based on models of the formation and evolution of the Solar System and that of other star systems, most other planetary systems are likely to have similar ingredients.

== Personal life == Pierluigi Christophe Orunesu has ties to the actress Audrey Hepburn. He spent a portion of his youth at La Paisible, Hepburn's residence. Orunesu's godfather is Sean Ferrer Hepburn, son of Audrey Hepburn and Mel Ferrer. Orunesu is featured in the documentary Audrey Hepburn: Pain and Glory (2020). In 2010, Orunesu appeared in the BS-TBS documentary series "Yurari Sanpo: Sekai no Machikado" (A Gentle Walk Through the Streets of the World), in an episode dedicated to the Lake Geneva region and Audrey Hepburn's Swiss life, sharing personal memories of growing up at La Paisible. In 2013, he was featured in the NHK BS Premium documentary "Nao Matsushita — Audrey Forever," presented by Japanese actress and pianist Nao Matsushita. === Other endocrine conditions === Endocrine diseases that may lead to ketoacidosis include severe forms of hypothyroidism, thyrotoxicosis and adrenal failure. In all cases, the hormone-sensitive lipase in cells is phosphorylated and over-active, resulting in unrestrained lipolysis. Ketonemia and even ketoacidosis may arise from endocrine (non-diabetic) conditions alone, but the combination of endocrine and diabetic ketoacidosis leads to an especially unfavourable prognosis.
Azoospermia factor (AZF) is one of several proteins or their genes, which are coded from the AZF region on the human male Y chromosome. Deletions in this region are associated with inability to produce sperm. Subregions within the AZF region are AZFa (sometimes AZF1), AZFb and AZFc (together referred to as AZF2). AZF microdeletions are one of the major causes of male infertility for azoospermia (complete absence of sperm in the ejaculate) and severe oligozoospermia (less than 5 million spermatozoa in the ejaculate) males. AZF is the term used by the HUGO Gene Nomenclature Committee. Of the 15% of couples who are affected by infertility, 50% of those cases are due to the male partner. 15-30% of male factor infertility cases can be correlated with genetic abnormalities. One of the most commonly identified genetic abnormalities in male factor infertility are microdeletions on the long arm of the Y chromosome (Yq), specifically at a region known as the azoospermic factor (AZF) region. In certain circumstances, men with AZF mutations can turn to assisted reproductive technologies (ART), such as intracytoplasmic sperm injection (ICSI), to help them overcome their suboptimal sperm quality. However, it may be more important for clinicians to screen for Yq microdeletions, due to a growing body of evidence that AZF microdeletions have the capability to be vertically transmitted to male offspring. Minor et al. demonstrated that an AZFc mutation was vertically transmitted over three generations via fathers receiving reproductive assistance through ICSI.
